A DOT consortium pools safety-sensitive employees from multiple employers into a shared random testing program managed by a third-party administrator. Testing Methods Used in the DOT Random Drug & Alcohol Testing Program

Urine specimen collection is the federally approved method for DOT drug testing under 49 CFR Part 40. The standard DOT drug test screens for five substance categories: marijuana metabolites, cocaine metabolites, amphetamines (including methamphetamine), opioids, and phencyclidine (PCP). All specimens are analyzed by SAMHSA-certified laboratories, and non-negative results undergo confirmatory GC/MS testing before the Medical Review Officer (MRO) makes a final determination.

A Medical Review Officer reviews all non-negative drug test results before they reach the employer, protecting employees from false positives caused by legitimate prescription use.

DOT regulations require employers to retain drug and alcohol testing records for specified periods. Records of verified positive results, alcohol concentrations of 0.02 or greater, and refusals must be kept for a minimum of five years; records of negative and cancelled results are kept for one year. Since January 2020, all DOT drug and alcohol program violations and return-to-duty completions for FMCSA-covered drivers must be reported to the FMCSA Drug and Alcohol Clearinghouse. Employers are also required to query the Clearinghouse before hiring CDL drivers in safety-sensitive positions.

Other DOT Testing Types That Work Alongside the Random Program

Companies with multiple safety-sensitive employees must also maintain a written DOT drug and alcohol policy and ensure at least one supervisor has completed a reasonable suspicion supervisor training program.
